Disminució de la glucòlisi com a empremta metabòlica en les cèl·lules endotelials en la hipertensió pulmonar tromboembòlica crònica

CAT: La hipertensió pulmonar tromboembòlica crònica (CTEPH) es produeix a conseqüència de trombus no resolts que obstrueixen les artèries pulmonars i produeixen vasculopatia perifèrica en arteries no obstruïdes. Partint de la hipòtesi que una desregulació metabòlica en les cèl·lules endotelials (EC) contribueix a canvis cel·lulars que promouen un estretament vascular en CTEPH, aquest estudi es basa en caracteritzar l'empremta metabòlica de les EC-CTEPH. Mitjançant l'aïllament de les EC de pacients que han estat sotmesos a una enderectomia pulmonar i fent servir EC d'arteria pulmonar humanes (HPAE) de voluntaris sans com a controls. Els nivells d'expressió dels enzims metabòlics han estat determinats a partir del mRNA i els nivells de proteïna. Comparant amb HPAE, les EC-CTEPH presenten uns nivells inferiors d'HK-2 i LDH-A, que representen el primer i l'últim pas del procés de la glucòlisi. Els nivells de mRNA dels reguladors de la glucòlisi PFK-2/fructosa-2,6 bifosofatasa-3 i PFK-1 són més baixos en les EC-CTEPH. A nivell de transcripció el PDK1, l'inhibidor del PDH i el GDH-1, involucrats en el metabolisme de la glutamina, es troben també en nivells inferiors. L'oxidació dels àcids grassos i la via del pentosa fosfat no difereixen entre EC-CETPH i HPAE. Els resultats de l'article publicat recentment a la revista American Journal of Respiratory Cell and Molecular Biology liderat per la Dra. Olga Tura del Grup Respiratori de l'IDIBGI mostren una reducció del metabolisme de la glucòlisi de les EC de les arteries pulmonars en pacients amb CTEPH, fet que pot contribuir a la disfunció endotelial i una remodelació vascular en CTEPH.

ENG: Decreased glycolysis as metabolic fingerprint of endothelial cells in chronic2 thromboembolic pulmonary hypertension

Chronic thromboembolic pulmonary hypertension (CTEPH) is caused by non-resolved thrombi obliterating pulmonary arteries and peripheral vasculopathy in nonobstructed arteries. We hypothesized that metabolic dysregulation of endothelial cells (EC) contributes to cellular changes that promote vascular narrowing in CTEPH. The study aimed to characterize the metabolic fingerprint of EC-CTEPH. We used EC isolated from specimens removed at pulmonary endarterectomy. Human pulmonary artery endothelial (HPAE) cells from healthy donors were used as control cells. Expression levels of metabolic enzymes were studied at mRNA and protein levels. Compared to HPAE, EC-CTEPH showed lower mRNA and protein levels of hexokinase-2 and lactate dehydrogenase-A, which represent the first and last steps of the glycolytic process. mRNA levels of the glycolytic regulators 6-phosphofructo-2-kinase/fructose-2, 6-biphosphatase-3 and phosphofructokinase-1 were reduced in EC-CTEPH. Transcript levels of pyruvate dehydrogenase kinase-1 (PDK1), inhibitor of pyruvate dehydrogenase, and glutamate dehydrogenase-1, involved in glutamine metabolism, were also downregulated in EC-CTEPH. Fatty acid oxidation (FAO) and the pentose phosphate pathway (PPP) did not differ between EC-CTEPH and HPAE. The results of the article recently published in the American Journal of Respiratory Cell and Molecular Biology led by Dr. Olga Tura of the IDIBGI's Respiratory Group show a reduction in the metabolism of glycolysis of the pulmonary arteries in patients with CTEPH, which may contribute to endothelial dysfunction and vascular remodelling in CTEPH.
